Specimen Preparation:

Embryos were fixed overnight in 4% paraformaldehyde and washed three times with copious amounts of PBS. Embryos were stored in PBS + 0.5% Sodium Azide. Prior to imaging, the specimens were washed in PBS and placed into Fomblin. Excess PBS was carefully removed by mouth pipet.
